The N12 Treasure Route, an integral segment of the N12 National Route, spans from Emalahleni (Witbank) in Mpumalanga, passing through Delmas into Gauteng, traversing Johannesburg to Carletonville, and continuing through Soweto into the North West Province near the Vredefort Dome World Heritage Site. From there, it proceeds through the City of Matlosane/KOSH (Klerksdorp, Orkney, Stilfontein, and Hartbeesfontein), touching Wolmaransstad, Bloemhof, and Christiana, before reaching Kimberley in the Northern Cape Province.

The route extends through Hopetown, Strybenburg, and Britstown to the historic town of Victoria West. Just south of Victoria West, at Three Sisters, it merges with the N1 National Route, leading to Beaufort West. Continuing south through Derust and Dysseldorp, it moves on to Oudtshoorn and concludes its journey over the Outeniqua Pass, meeting the N2 National Route in George, near Victoria Bay and the Indian Ocean.

It is noteworthy that National Roads N1, N2, N3, and N4 intersect or share connections with the N12 National Road at various points along the route.
